A step closer to artificial livers: Researchers identify compounds that help liver cells grow outside body

June 2, 2013 ? Prometheus, the mythological figure who stole fire from the gods, was punished for this theft by being bound to a rock. Each day, an eagle swept down and fed on his liver, which then grew back to be eaten again the next day.

Modern scientists know there is a grain of truth to the tale, says MIT engineer Sangeeta Bhatia: The liver can indeed regenerate itself if part of it is removed. However, researchers trying to exploit that ability in hopes of producing artificial liver tissue for transplantation have repeatedly been stymied: Mature liver cells, known as hepatocytes, quickly lose their normal function when removed from the body.

"It's a paradox because we know liver cells are capable of growing, but somehow we can't get them to grow" outside the body, says Bhatia, the John and Dorothy Wilson Professor of Health Sciences and Technology and Electrical Engineering and Computer Science at MIT, a senior associate member of the Broad Institute and a member of MIT's Koch Institute for Integrative Cancer Research and Institute for Medical Engineering and Science.

Now, Bhatia and colleagues have taken a step toward that goal. In a paper appearing in the June 2 issue of Nature Chemical Biology, they have identified a dozen chemical compounds that can help liver cells not only maintain their normal function while grown in a lab dish, but also multiply to produce new tissue.

Cells grown this way could help researchers develop engineered tissue to treat many of the 500 million people suffering from chronic liver diseases such as hepatitis C, according to the researchers.

Lead author of the paper is Jing (Meghan) Shan, a graduate student in the Harvard-MIT Division of Health Sciences and Technology. Members of Bhatia's lab collaborated with researchers from the Broad Institute, Harvard Medical School and the University of Wisconsin.

Bhatia has previously developed a way to temporarily maintain normal liver-cell function after those cells are removed from the body, by precisely intermingling them with mouse fibroblast cells. For this study, funded by the National Institutes of Health and Howard Hughes Medical Institute, the research team adapted the system so that the liver cells could grow, in layers with the fibroblast cells, in small depressions in a lab dish. This allowed the researchers to perform large-scale, rapid studies of how 12,500 different chemicals affect liver-cell growth and function.

The liver has about 500 functions, divided into four general categories: drug detoxification, energy metabolism, protein synthesis and bile production. David Thomas, an associate researcher working with Todd Golub at the Broad Institute, measured expression levels of 83 liver enzymes representing some of the most finicky functions to maintain.

After screening thousands of liver cells from eight different tissue donors, the researchers identified 12 compounds that helped the cells maintain those functions, promoted liver cell division, or both.

Two of those compounds seemed to work especially well in cells from younger donors, so the researchers -- including Robert Schwartz, an IMES postdoc, and Stephen Duncan, a professor of human and molecular genetics at the University of Wisconsin -- also tested them in liver cells generated from induced pluripotent stem cells (iPSCs). Scientists have tried to create hepatocytes from iPSCs before, but such cells don't usually reach a fully mature state. However, when treated with those two compounds, the cells matured more completely.

Bhatia and her team wonder whether these compounds might launch a universal maturation program that could influence other types of cells as well. Other researchers are now testing them in a variety of cell types generated from iPSCs.

In future studies, the MIT team plans to embed the treated liver cells on polymer tissue scaffolds and implant them in mice, to test whether they could be used as replacement liver tissues. They are also pursuing the possibility of developing the compounds as drugs to help regenerate patients' own liver tissues, working with Trista North and Wolfram Goessling of Harvard Medical School.

Eric Lagasse, an associate professor of pathology at the University of Pittsburgh, says the findings represent a promising approach to overcoming the difficulties scientists have encountered in growing liver cells outside of the body. "Finding a way of growing functional hepatocytes in cell culture would be a major breakthrough," says Lagasse, who was not part of the research team.

Making connections

Bhatia and colleagues have also recently made progress toward solving another challenge of engineering liver tissue, which is getting the recipient's body to grow blood vessels to supply the new tissue with oxygen and nutrients. In a paper published in the Proceedings of the National Academy of Sciences in April, Bhatia and Christopher Chen, a professor at the University of Pennsylvania, showed that if preformed cords of endothelial cells are embedded into the tissue, they will rapidly grow into arrays of blood vessels after the tissue is implanted.

To achieve this, Kelly Stevens in the Bhatia lab worked with Peter Zandstra at the University of Toronto to design a new system that allows them to create 3-D engineered tissue and precisely control the placement of different cell types within the tissue. This approach, described in the journal Nature Communications in May, allows the engineered tissue to function better with the host tissue.

"Together, these papers offer a path forward to solve two of the longstanding challenges in liver tissue engineering -- growing a large supply of liver cells outside the body and getting the tissues to graft to the transplant recipient," Bhatia says.

The rise of Massive Open Online Courses is presenting higher education with a powerful challenge. Access to great teachers will help millions. But will MOOCs cause a massive college shakeout as well?

A tree farm produces a monoculture you can count on. Its timber efficiently becomes the lumber that makes houses and furniture. A woodlot is a little more sketchy. It might begin as a forgotten weed patch, grow into a scrubby forest, and eventually host a mini-United Nations of species. Left alone, a woodlot can become an interestingly varied patch of earth, maybe even a natural treasure.

Conventionality or originality? Most of us choose both. We don?t want surprises when it comes to floor joists. We prefer our airline pilots not to let the muse guide them to Pittsburgh. But leave room for serendipity. Order keeps our world humming. The unthought-of tips the world?s equilibrium. It can be as disruptive as quantum physics, as fresh as Beethoven or The Beatles.

Education is forever balancing and rebalancing uniformity and creativity. Basic competence has to be mastered. But innovative thinking must be encouraged. Read the canon of great literature, but don?t be afraid to demolish conventional wisdom. Students and their parents seek out the best school and best teachers, hoping for the best education. But students can flourish at middling colleges and with average teachers if their reading is inspiring, their lab work intriguing, their thinking encouraged.

When you read Laura Pappano?s cover story on the huge stir being caused by Massive Open Online Courses (MOOCs, pronounced ?mooks?), you may at first think that there?s nothing new under the sun. Correspondence courses, after all, began in the 19th century. Over the decades, educational institutions have experimented with teaching via radio, television, closed-circuit video, and the Internet. And each new distance-learning technology has prompted predictions of the demise of ivy-clad campuses, the loss of mentoring by belovedly quirky profs, and the end of fond memories of college life. Fifteen years ago, a reporter from The Boston Globe marveled at how 1990s cutting-edge technology ? ?a two-way PictureTel compressed-video system linked by high-speed phone lines? ? was connecting a classroom on Martha?s Vineyard with a university on the Massachusetts mainland. As one university official told him (well, actually, told me): ?What is better in terms of quality ? a dull, boring, standard lecture, or a penetrating lecture by a great teacher, backed up with all the best video props...??

The PictureTel wonderment didn?t disrupt the college paradigm back then. Will MOOCs? Perhaps. The technology and pedagogy of online ed is constantly improving. And the pressing need to control costs seems destined to drive online education forward. That worries some people. This spring, philosophy professors at San Jose State University in California sent a protest letter to political philosophy superstar Michael Sandel of Harvard University decrying the MOOCing of his course of social justice. Among other things, they warned, ?the thought of the exact same social justice course being taught in various philosophy departments across the country is downright scary ? something out of a dystopian novel.?

A balance needs to be struck between the franchising of high-quality education and the more intimate, locally grown experience that occurs when teachers and students reason together in a classroom. It seems inevitable that the MOOC monoculture will spread. But let?s make sure we preserve the woodlot. Amazing, unthought-of ideas could be growing in it.?

Pistorius lawyer demands answers over bloody leaked pictures

JOHANNESBURG (Reuters) - Lawyers for "Blade Runner" Oscar Pistorius demanded answers from South African police and prosecutors on Friday after pictures of the bathroom and toilet where he shot dead his girlfriend were leaked to a British television station.

Sky News obtained a series of images of the crime scene in Pistorius' luxury Pretoria home where the double amputee track star shot his girlfriend, model Reeva Steenkamp, on Valentine's Day on February 14.

Pistorius was one of the stars of the 2012 Olympics and Paralympics in London and the case shocked millions across the world as he was seen as a symbol of truimph over adversity.

In South Africa, he was a hero for both blacks and whites, transcending the racial divides that persist 19 years after the end of apartheid.

Pistorius has admitted to shooting four times through the locked door but says he was acting in self-defense at what he thought was an intruder.

Prosecutors accuse him of premeditated murder for firing into the door hitting Steenkamp, 29, in the head, hip and arm.

The pictures showed a pool of blood in the bathroom, spattered walls, and a trail from dragging the body.

Lawyer Brian Webber said he had sent a letter to the prosecution team demanding answers for the appearance of the pictures the week before Pistorius is due back in court.

"I have requested an explanation and I am confident I will get one," he told Reuters.

A spokesman for the National Prosecuting Authority said it was unclear how Sky had obtained the pictures. Police declined to say whether they were part of the formal investigation but said they would make no difference to the trial.

South Africa does not have trial by jury so pre-trial media coverage is seldom deemed to be prejudicial.

Tuesday's court appearance by Pistorius is expected to be a 10-minute formality.

Canadian writer-director Kim Nguyen spent nearly a decade researching this docudrama about child soldiers in Africa, and the film feels as authoritative as a first-hand account.

A haunting take on unspeakably grim subject matter, shot on location in the Democratic Republic of Congo.

A powerful and upsetting portrait of a young girl compelled into unimaginably horrific circumstances.

Nguyen, astonishingly, manages to wring something vaguely like a happy ending from this tragic story.

War Witch is most effective not when we are looking in on Komona but when we are inside her head.

The powerful things we expect from "War Witch" are as advertised, but what we don't expect is even better.

... driven by a remarkably natural, unaffected performance by Mwanza. And Nguyen, despite relying a little too heavily on the initial voice-over for exposition, is a confident and sensitive intelligence behind the camera.

You're likely to ponder its images, its insights into a very foreign (for most of us) location and the tragic situation of Komona (and others like her) for a long time to come.

Is it accurate depiction of Africa's child soldiers? I don't know, thank God. But it feels authentic to its very core, and that makes it as hard to forget as it is to ignore.

Brutal without turning exploitative, the result is harrowing and heartbreaking.

Nguyen creates a mesmerizing tone through his camerawork, editing, sound and the infusion of African folk imagery and ritual, but it's Mwanza's performance as Komona that makes "War Witch" feel so miraculous.

Nguyen reportedly worked on "War Witch" for a decade, and it shows in both the immediacy and authenticity of his tale, and the meticulous craft with which it's told.

Made with extremely clear-eyed restraint from harangues, sentiment, message-mongering, or anything else that would cheapen its central character's suffering and fight.

War Witch features a standout performance by Rachel Mwanza, but the supernatural visions don't really suit the film's tone and mood.

Nguyen's compassion and commitment to the issue is admirable, and at its best, War Witch is devastating.

War Witch is remarkable for the fact that it never strays into sentimentality or sensationalism.

...a love story between youngsters who are forced to become adults all too early in their lives.

This is a straight ahead essay on warfare at its worst and the survival of the human spirit at its best.

An astonishing drama set in Africa that vividly depicts the courage and resiliency of a 12-year-old girl whose spiritual gifts enable her to survive.

It is astonishing that film that contains such violence can have such a serene tone. The source of the serenity is the measured, calm narration by Komona (voice of Diane Umawahoro) that is the telling of her story to her unborn child

An exquisitely made film in direct contrast to the ugliness of its subject matter

Remapping Our Awareness Of Storm Surge Danger

Hurricane season begins Saturday, and the National Oceanic and Atmospheric Administration (NOAA) is forecasting an active season, with perhaps 7 to 11 hurricanes.

With memories of last year's destruction from Hurricane Sandy still fresh, meteorologists are working on ways to improve the way they forecast storms and communicate warnings to the public.

When Sandy was making its way northward in the Atlantic and began to turn toward the East Coast, the National Hurricane Center tried to emphasize the danger that storm surge posed for residents, especially those near New York City.

The Hurricane Center's forecast warned of six to eleven feet of storm surge. As it turned out, they were right on the money. Rising seawater, pushed by Sandy's powerful winds, came ashore and brought widespread flooding to New Jersey, Long Island, parts of Brooklyn and Staten Island.

"It was like the ocean came up on land," says resident Rosemary Caruso, who lives just a block from the water on Staten Island.

Underestimating Storm Surge

This wasn't the first time a storm had flooded Staten Island. There were storm surges in the 1930s, but back then many of the coastal areas were still undeveloped marshland.

When Sandy hit, thousands in vulnerable neighborhoods didn't evacuate, despite the warnings. More than 20 people died on Staten Island, most from drowning. Jamie Rhome, a storm surge specialist at the National Hurricane Center, says it's a problem seen in nearly every hurricane.

"In general, this nation does not understand storm surge. Nor does it understand its vulnerability," Rhome says. "We've seen poll after poll of individuals, right at the coast, saying they're not vulnerable to storm surge."

As Sandy showed, it's a threat that's not limited to states along the Gulf and in the southeast.

"The New York area is, what I think, one of the most vulnerable areas in the entire East Coast," says William Fritz, a geologist and president of the College of Staten Island.

Months before Sandy, Fritz worked with Alan Benimoff to map areas on the island vulnerable to storm surge. The map proved prescient. The morning after the storm, Benimoff was out on Staten Island charting the surge.

"It was terrible. I never saw anything like it," Benimoff says.

In some neighborhoods, he says, Sandy's storm surge reached 14 feet. It matched Fritz and Benimoff's model almost exactly.

With the help of supercomputers at the University of North Carolina, Fritz says his team is now prepared to do real-time modeling to understand the threat of storm surge as a hurricane approaches.

The problem, he says, is that public awareness of the threat of storm surge now lags behind science's ability to forecast it.

The National Hurricane Center is working on new storm surge maps ? like this one of Miami-Dade County in Florida ? to help residents understand their risk.

"Some of the people that lost their lives ... were a few blocks away from safe ground," Fritz says. "But they didn't know ... where the safe ground was."

Fritz is calling for a public education campaign similar to that used in the northwestern U.S. for tsunamis; one that would include signs identifying surge-prone areas and the nearest high ground.

Remapping The Threat

Learning from the experience of Sandy, Florida's Miami-Dade County has recently drawn up new maps dividing the county into five evacuation areas now relabeled "storm surge zones."

The new maps have tripled the number of people who live in areas that could potentially be evacuated. Emergency Management director Curt Sommerhoff says that's nearly two million residents ? most of the people who live in Miami-Dade County.

"We're in the most hurricane-prone area of the most hurricane-prone state in the United States," Sommerhoff says. Since Florida is a peninsula, he says, it faces storm surge threats on three different fronts.

In a hurricane, emergency managers advise residents to shelter in place when at risk from high winds. But when there's a risk of storm surge ? which causes the most damage and takes the most lives ? they tell residents to evacuate.

Efforts to get that message to the public are also underway at the National Hurricane Center. One of the new tools is graphics and mapping software designed to be read and easily understood by the public.

Instead of technical terms like "height above mean sea level," the map tells coastal residents how deep the water is likely to be in their neighborhoods. The map is color-coded, too, explains the Center's Jamie Rhome, who helped design the software.

"The brighter the color, the deeper the water has the potential to be," Rhome says. "But more importantly, people don't understand how far the water goes inland. This map clearly shows how far the water goes inland. If you're in one of those colored areas, you need to do something to protect your life."

The new storm surge maps were intended to be ready by next year, National Hurricane Center officials say, but because of the need and strong demand by emergency managers, they hope to begin using them this summer.
