Hundreds of alterations and potential drug targets to starve tumors identified

A massive study analyzing gene expression data from 22 tumor types has identified multiple metabolic expression changes associated with cancer. The analysis, conducted by researchers at Columbia University Medical Center, also identified hundreds of potential drug targets that could cut off a tumor's fuel supply or interfere with its ability to synthesize essential building blocks. The study was published today in the online edition of Nature Biotechnology.

The results should ramp up research into drugs that interfere with cancer metabolism, a field that dominated cancer research in the early 20th century and has recently undergone a renaissance.

"The importance of this new study is its scope," said Dennis Vitkup, PhD, associate professor of biomedical informatics (in the Initiative in Systems Biology) at CUMC, the study's lead investigator. "So far, people have focused mainly on a few genes involved in major metabolic processes. Our study provides a comprehensive, global view of diverse metabolic alterations at the level of gene expression."

Cell metabolism is a dynamic network of reactions inside cells that process nutrients, such as glucose, to obtain energy and synthesize building blocks needed to produce new cellular components. To support uncontrolled proliferation, cancer needs to significantly reprogram and "supercharge" a cell's normal metabolic pathways.

The first researcher to notice cancer's special metabolism was German biochemist Otto Warburg, who in 1924 observed that cancer cells had a peculiar way of utilizing glucose to make energy for the cell. "Although a list of biochemical pathways in normal cells was comprehensively mapped during the last century," said Dr. Vitkup. "We still lack a complete understanding of their usage, regulation, and reprogramming in cancer."

"Right now we have something like a static road map. We know where the streets are, but we don't know how traffic flows through the streets and intersections," said Jie Hu, PhD, a postdoctoral researcher at Columbia and first author of the study. "What researchers need is something similar to Google Traffic, which shows the flow and dynamic changes in car traffic."

Drs. Hu and Vitkup's study is an important step toward achieving this dynamic view of cancer metabolism. Notably, the researchers found that the tumor-induced expression changes are significantly different across diverse tumors. Although some metabolic changes?such as an increase in nucleotide biosynthesis and glycolysis?appear to be more frequent across tumors, others, such as changes in oxidation phosphorylation, are heterogeneous.

"Our study clearly demonstrates that there are no single and universal changes in cancer metabolism," said Matthew Vander Heiden, MD, PhD, assistant professor at MIT, and a co-author of the paper. "That means that to understand transformation in cancer metabolism, researchers will need to consider how different tumor types adapt their metabolism to meet their specific needs."

The researchers also found that expression changes can mimic or cooperate with cancer mutations to drive tumor formation. A notable example is the enzyme isocitrate dehydrogenase. In several cancers, such as glioblastoma and acute myeloid leukemia, mutations in this enzyme are known to produce a specific metabolite?2-hydroxyglutarate?that promotes tumor growth. The Columbia team found that isocitrate dehydrogenase expression significantly increases in tumors with the recurrent mutations. Such an overexpression may create an efficient enzymatic factory for overproduction of 2-hydroxyglutarate.

The analysis also led the researchers to an interesting finding in colon cancer. In several other cancers, mutations in two enzymes?succinate dehydrogenase and fumarate hydratase?can promote tumor formation as a result of efflux from mitochondria and accumulation of their substrates, fumarate and succinate. The researchers found that in colon cancer, accumulation of these metabolites may be caused by a significant decrease in the enzymes' expression. This was confirmed when metabolomics data from colon tumor patients showed significantly higher concentrations of fumarate in tumors than in normal tissue.

"These are just several examples of how cancer cells use various creative mechanisms to hijack the metabolism of native cells for their own purposes," said Dr. Vitkup.

For cancer researchers looking for new drug targets, Dr. Vitkup's team also found hundreds of differences between normal and cancer cells' use of isoenzymes. This opens up additional possibilities for turning off cancer's fuel and supply lines. Isoenzymes often catalyze the same reactions, but have different kinetic properties: Some act quickly and sustain rapid growth, while others are more sluggish. In kidney and liver cancers, for example, a quick-acting aldolase isoenzyme?suitable for fast cell proliferation?was found to be more prevalent than the more typical slow-moving version found in normal kidney and liver tissue. Although a few examples of differential isoenzyme expression in tumors were already known, the Columbia researchers identified hundreds of isoenzymes with cancer-specific expression patterns.

"Inhibiting specific isoenzymes in tumors may be a way to selectively hit cancer cells without affecting normal cells, which could get by with other isoenzymes," said Dr. Hu.

In fact, a recent study from Matthew Vander Heiden's laboratory demonstrated the potential of targeting a specific isoenzyme, pyruvate kinase M2, expression of which often increases in tumors. "The comprehensive expression analysis suggests that a similar approach could potentially be applied in multiple other cases," said Dr. Vander Heiden.

Targeting metabolism may be a way to strike cancer at its roots. "Cancer cells usually have multiple ways to turn on their growth program," said Dr. Vitkup. "You can knock out one, but the cells will usually find another pathway to turn on proliferation. Targeting metabolism may be more powerful, because if you starve a cell of energy or materials, it has nowhere to go."

If all goes well, the Antares rocket, built and operated by Orbital Sciences Corp., will join Space Exploration Technologies Corp.'s (SpaceX) Falcon 9 as replacements for the space shuttles that carried cargo to and from the ISS. The Falcon 9 and its Dragon cargo capsule already have completed two formal resupply missions to the station.

"We did come late to this activity," acknowledges Frank Culberston, a former NASA astronaut and now an executive vice president at Orbital Sciences. The company signed on to NASA's Commercial Orbital Transportation Services (COTS) program about 1-1/2 years after the program began, after NASA dropped one of the two initial participants.

"We've been playing catch-up, but we've about caught up. By the end of next year, we should have another four or five cargo missions under our belt," he adds.

NASA's initial choice of two companies for the COTS program involved aerospace upstarts born within a year of each other. Now, the program has paired a grizzled spaceflight veteran with a precocious tweenager.

Orbital Sciences has been building satellites and building and launching rockets for more than 30 years. SpaceX was founded in 2002.

Their approaches to designing and building their rockets are markedly different.

SpaceX has prided itself on designing and building its hardware in-house, beginning with its two-stage, liquid-fueled Falcon 1 rocket. The Falcon 1, with a single engine in its first stage, had teething problems. First launched in March 2006, the rocket's initial three missions failed to deliver anything to orbit. After a fourth, successful test that put a dummy payload into low-earth orbit, the Falcon 1 went on to loft a Malaysian remote-sensing satellite a year later, only to be retired as SpaceX focused on its workhorse, the Falcon 9.

Nearly 200 feet tall, the Falcon 9 boasts nine main engines and can loft much heavier payloads than its predecessor. Sometime this year, the company plans to test a third rocket, the Falcon Heavy ? in effect three Falcon 9s strapped side by side, with the center booster topped with a second stage and a payload. The company bills the Falcon Heavy as the most powerful rocket the US has seen since the Apollo program's Saturn V rocket in the 1960s and '70s.

The company's ultimate quest is to develop reusable rockets, in addition to the reusable capsule it has built to carry cargo and eventually humans. SpaceX is testing a propulsion system that will allow a rocket booster to land vertically.

G-20 officials decry lack of global growth

WASHINGTON (AP) ? World finance leaders say they are determined to attack a sluggish global economy in which growth is too weak and unemployment too high. Their problem is arriving at a consensus over the proper mix of policies.

Finance ministers and central bank presidents from the world's biggest economies issued a joint statement Friday that papered over stark differences between opposing views. The United States and other countries are pushing for less budget austerity and more government stimulus while Germany and others contend that attacking huge budget deficits should be job No. 1.

The discussions were scheduled to wrap up Saturday with meetings of the steering committees of the 188-nation International Monetary Fund and its sister lending agency, the World Bank.

The G-20 joint statement revealed no major new policy initiatives and sought to straddle the divide in the growth-and-austerity argument.

The United States is being represented at the talks by Treasury Secretary Jacob Lew and Federal Reserve Chairman Ben Bernanke.

"Strengthening global demand is imperative and must be at the top of our agenda," Lew said in remarks late Friday before the IMF policy-setting group. "Stronger demand in Europe is critical to global growth."

However, other nations, led by Germany, have resisted a move away from austerity programs, saying they are critical to getting government deficits under control.

German Finance Minister Wolfgang Schaeuble apologized to a Washington audience for being late for a speech after the G-20 discussions Friday, saying, "On reduction of indebtedness ... we have a little bit of differences of opinion all over the world, to be very frank, and that's the reason I am a little bit late."

Dutch Finance Minister Jeroen Dijsselbloem, the head of the Eurogroup, encompassing the 17 finance ministers whose countries use the euro currency, said that European nations needed to keep pushing to reduce huge budget deficits but "we can and will adjust" the speed that the deficit cuts are implemented to take into account economic conditions.

The G-20 joint statement singled out the recent aggressive credit-easing moves pushed by Japanese Prime Minister Shinzo Abe, saying they were intended to stop prolonged deflation and support domestic demand.

Those comments were viewed as giving a green light to Japan's program, which has driven the value of the yen down by more than 20 percent against the dollar since October. That sizable decline has raised concerns among U.S. manufacturing companies that Japan's real goal is not to fight deflation, a destabilizing period of falling prices, but to weaken the yen as a way to gain trade advantages.

To address those concerns, the G-20 did repeat language it used in February that all countries should not use their currency as a trade weapon and guard against policies that could trigger currency wars.

Japanese officials told reporters following the discussions that they were pleased by the support the G-20 had given them to pursue growth policies in an effort to lift the world's third-largest economy out of its two-decade slump.

Haruhiko Kuroda, head of the Bank of Japan, said, "There has been international understanding and acceptance of this, so we can have further confidence to appropriately conduct monetary policy."

The G-20 statement also said that there was an urgent need for the euro currency area to move toward a banking union and reduce the "financial fragmentation" that now exists.

The communique said that "more needs to be done to address the issues of international tax avoidance and evasion in particular through havens." The financial crisis that hit the Mediterranean island of Cyprus earlier this year revived concern over countries that serve as tax havens.

In Cyprus, banks held more than $162 billion in assets, or roughly seven times the country's total GDP. Much of that money came from wealthy Russian investors.

Labor Nominee Faces GOP Scrutiny

Secretary of Labor nominee Thomas Perez faced two hours of intense scrutiny at his nomination hearing Thursday morning as he sought to alleviate Republican concerns over his role in the Civil Rights Division of the Department of Justice.

At issue is a lengthy report released earlier this week in which GOP leaders accuse Perez of attempting to influence the city of St. Paul, Minn., to withdraw a housing discrimination case before it could be brought before the Supreme Court. In exchange, the Department of Justice agreed not to intervene in two whistleblower cases against St. Paul that could have won up to $200 million for taxpayers.

Sen. Lamar Alexander engaged Perez in a heated line of questioning, accusing Perez of doing ?an extraordinary amount of wheeling and dealing.?

?You have a duty to protect the money, a duty to protect the whistleblower, and at the same time, it seems to me that you?re manipulating the legal process to try to get the result you want from the Supreme Court in a way that?s inappropriate,? Alexander, R-Tenn., said.

Perez defended his actions, saying DOJ chooses not to stay out of ?a lot of different things.?

?It was in the interest of justice and it was entirely appropriate to do so in the opinion of professional responsibility people and others,? he told the Senate Health, Education, Labor, and Pensions Committee. ?I believe the resolution reached in this case was in fact in the interest of justice.?

Democratic senators had kind words for Perez. Sen. Barbara Mikulski, D-Md., introduced him as ?one of Maryland?s favorite sons,? saying ?we believe he is the right man for the job.?

Senator Patty Murray, D-Wash., joined in the praise, adding ?you have a very amazing, impressive, wide range of experience that you are bringing from a number of different agencies?You?re something of a turn-around expert for public sector agencies, so thank you for that.?

When asked what his very top priority would be should he be confirmed as Secretary of Labor, Perez had one answer: ?jobs, jobs, jobs.?

He expanded on his goals and priorities, including reauthorizing the Workforce Investment Act, maintaining pension security, and spending his first 100 days as secretary on a listening tour of America, reaching out to small businesses and workers alike.

?The president has asked all of us to consider three questions in the decisions we make,? Perez said. ?How do we make America a magnet for jobs? How do we equip our people with the skills they need to succeed in those jobs, and how do we ensure that an honest day?s work leads to a decent living??

Perez concluded, ?These questions are at the core of the mission of the Department of Labor, and if confirmed you have my word that I will keep them there.?

The committee is expected to vote on Perez?s nomination on Thursday, April 25.
